What Is FOXO4-DRI?
FOXO4-DRI is a synthetic D-retro-inverso peptide designed to disrupt the interaction between the FOXO4 transcription factor and p53, which selectively pushes senescent (“zombie”) cells into apoptosis while sparing normal cells. It is a proof-of-concept senolytic tool compound from aging research, not an approved dietary supplement or medicine, and its evidence base is preclinical.
Evidence
The defining study designed the FOXO4 peptide, showed it causes p53 nuclear exclusion and cell-intrinsic apoptosis specifically in senescent cells, and reported that in mice it neutralised chemotherapy-induced toxicity and restored fitness, fur density, and kidney function in both fast-aging and naturally aged animals 1Reference 1In vitroTargeted Apoptosis of Senescent Cells Restores Tissue Homeostasis in Response to Chemotoxicity and Aging — mouse / in vitroView study →. That is a cell and mouse study. There is no verified human clinical trial of FOXO4-DRI for aging or any indication that I could find in this quick pass — the “clears senescent cells / reverses aging in humans” framing is extrapolated from mouse data and should be treated as unproven in people.
Dosage & Safety
FOXO4-DRI is a research peptide — not an approved dietary supplement or medicine, with no established human dose and no human safety data. Because its mechanism deliberately triggers cell death, the risks of unsupervised human use are genuinely unknown. Interactions and use in pregnancy are not established and were not assessed in this pass. This entry is informational only and is not a recommendation to use.
