P21

P21 (P021) is a synthetic CNTF-derived research peptide studied in rodents for neurogenesis and cognitive rescue in Alzheimer's models; it is not an approved drug or supplement and has no human trials.

What Is P21?

P21 (also written P021) is a small synthetic peptide derived from the biologically active region of ciliary neurotrophic factor (CNTF), engineered to promote neurogenesis and neuroprotection. It was developed in Khalid Iqbal’s laboratory at the New York State Institute for Basic Research; the peptide carries an adamantylated modification to improve blood–brain-barrier penetration and resist breakdown. In research it is studied as a candidate treatment for cognitive decline, Alzheimer’s disease, and other neurodegenerative conditions. It is a research compound, not an approved medicine or dietary supplement, and there are no published human clinical trials.

Evidence

The evidence for P21 is entirely preclinical (rodent). These are consistent and encouraging animal findings, but they are animal findings: no human trial of P21 exists in this pass, so its efficacy and safety in people are unproven, and popular claims that it “boosts neurogenesis” in humans are not backed by a verified clinical study.

Dosage & Safety

There is no established human dose for P21 and no controlled human safety data; any regimens circulating online are extrapolated from animal work, and this page makes no dosing recommendation. P21 is not FDA-approved and is not an approved dietary supplement. Human safety, drug interactions, long-term effects, and pregnancy/lactation safety are not established and have not been assessed in this pass.
