PRL-8-53 is a synthetic research nootropic reported in a single 1970s human study to improve verbal memory retention; modern evidence is essentially absent.
What Is PRL-8-53?
PRL-8-53 is a synthetic compound (a benzoic-acid methyl-ester derivative) marketed as a research nootropic for memory enhancement. It is not an approved drug or dietary supplement and is sold as a research chemical. Its reputation rests almost entirely on a single small human study from 1978; there is no substantial modern clinical literature, so any claims about it should be read with strong caution.
Evidence
The only primary human trial is Hansl & Mead (1978), a double-blind, placebo-controlled study reporting that a low oral dose improved retention of verbal information (nonsense-syllable learning) to a statistically significant degree, with acquisition only slightly improved 1Reference 1RCTPRL-8-53: enhanced learning and subsequent retention in humans as a result of low oral doses of new psychotropic agent — randomised double-blind placebo-controlled trialView study →. This is a single, old, small study and has not, to my knowledge, been independently replicated in a modern controlled trial — I did not find any citable modern human RCT of PRL-8-53 in this pass. The compound’s toxicity, long-term safety, and pharmacokinetics in humans are not well characterised. Marketing and nootropic-community write-ups frequently amplify the 1978 result (e.g., “doubled recall”); those secondary summaries are not additional evidence and should not be treated as such.
