L-Leucine
L-Leucine is an essential branched-chain amino acid and the primary amino-acid trigger of the mTOR pathway that initiates muscle protein synthesis.
What Is L-Leucine?
L-Leucine is an essential branched-chain amino acid (BCAA) and the single most important amino-acid signal for muscle protein synthesis. Along with isoleucine and valine it forms the BCAA group, but leucine is the one with a distinct pharmacological action: it activates the mTOR pathway, which switches on the translation machinery that builds new muscle protein. Being essential, it can’t be made by the body and must come from diet — it is abundant in meat, dairy, eggs, soy and other complete proteins.
What It’s Used For
Leucine is sold on its own and as the headline ingredient of BCAA and essential-amino-acid (EAA) blends, aimed mainly at muscle growth, exercise recovery, and preserving lean mass — including in older adults with age-related muscle loss (sarcopenia). The honest picture is mixed: leucine reliably fires the synthesis signal, but on its own it can’t sustain new muscle protein without the other essential amino acids as building material, so results for isolated leucine are inconsistent. It’s most useful as part of a complete protein or full EAA source rather than in isolation.
How It Works
Rising blood leucine after a protein-containing meal acts as a nutrient-sensor signal: it activates mTORC1, which initiates the translation step of muscle protein synthesis. Leucine is also metabolised in muscle (rather than the liver) for energy, and can feed into ketone-body production. Its downstream metabolite HMB is sometimes supplemented separately for anti-catabolic effects.
Dosage
Studied leucine doses are typically around 2–3 g per serving — roughly the leucine “threshold” thought to maximise the synthesis trigger — often delivered within a 5–10 g/day BCAA blend at a 2:1:1 leucine:isoleucine:valine ratio. These figures are reported for context and are not a personal recommendation.
Safety
Leucine is an essential nutrient present in ordinary dietary protein and is well tolerated in healthy adults at food-level and typical supplemental intakes. Very high isolated BCAA intakes are an area of ongoing research (the branched-chain-amino-acid/insulin-sensitivity question). Specific drug interactions and safety in pregnancy, kidney or liver disease are pending the detailed research buildout; dietary intake from food is not a concern.
